Abstract
NEW MATERIAL:A cyclopropylmethylnormorphine-6-sulfuric ester shown by the formula I and its pharmaceutically acceptable salt.
USE: A narcotic antagonism analgesic. Better than nalorphine hydrochloride, having prolonged actions. Administration by injection is preferable.
PROCESS: A compound shown by the formula II is acetylated to give a compound shown by the formula III, which is formed into a sulfuric ester, to give a compound shown by the formula IV. This compound is then deacetylated to give a compound shown by the formula I. The acetylation is carried out by reacting the compound shown by the formula II with acetic anhydide, acetyl chloride, etc. in the presence of an aqueous solution of an alkali metal bicarbonate, e.g., NaHCO3, KHCO3, etc. The formation of the sulfuric ester is done by reacting the compound shown by the formula III with chlorosulfonic acid in the presence of pyridine, etc. THe deacetylation is carried out by reacting the compound shown by the formula IV with an alcoholic solution of NaOH, etc.
