Abstract
PURPOSE: To prepare the titled preparation exhibiting strong adhesivity in the local application to the microtissue, and effective for the remedy of wound, by freezing a concentrated solution of fibrinogen obtained by adding a monosaccharide to a buffer solution having low salt concentration, keeping the solution at a specific temperature, and dissolving dried fibrinogen to the solution.
CONSTITUTION: A monosaccharide (e.g. glucose, fructose, mannitol, etc.) is added to a buffer solution having low salt concentration and a pH of 5.5W7 (e.g. 0.01W 0.03M citric acid buffer solution) in an amount of 3W6 (W/V)%, and maintained at 20W30°C. Powder of dried fibrinogen is dissolved in the solution to obtain concentrated solution containing 6W10(W/V)% fibrinogen. The solution is disinfected, filtered, poured into ampules, and frozen to obtain the frozen fibrinogen preparation. The preparation is stored at ≤-22°C, and is locally applied together with thrombin, aprotinin, etc. after melting. Dose: 10W30cc of the frozen preparation containing 9% fibrinogen.
