I had chalked ESM3 up as just another AI hypetrain. But generating a protein by describing it using natural language is what makes this such a landmark achievement. This week I had the pleasure of writing about some awesome new releases in #synbio - ESM3 and the new CRISPR-less DNA manipulation technique. Both are extremely promising and honestly quite sci-fi. ESM3 being able to generalise and reason over the endless reams of biological sequence data to actually identify patterns, and navigate through them using natural language... it's like we've cracked the Rosetta Stone but for biology. It's hard for me to convey just how exciting I think that is. And CRISPR-less DNA manipulation... well, I mean, that's not huge in itself. TALENs and excision enzymes could already do this. The actual fascinating hook of bridge RNA recombinases is that it enables DNA manipulation *without breaking any DNA*. Which, if you think of genomes as essentially being LEGO sets whereby the bricks are the base pairs... well, it just makes no physical sense right? They key thing being that it uses covalent intermediates. Essentially, unlike CRISPR, it never leaves a broken strand exposed. It just swaps one bond for another. This was so much fun to write about, and I look forward to seeing how these tools end up shaping the future of the bioeconomy. It was a pleasure to attend the ESACT - European Society for Animal Cell Technology meeting a few weeks ago to share some exciting data we have generated at Oxford Biomedica in collaboration with Sartorius and UCL Biochemical Engineering. Through the use of a novel anion-exchange membrane that has been designed specifically for Lentiviral Vector purification, we have been able to obtain a 3-fold boost in LV recovery over the downstream process. With this novel membrane, DSP LV recovery as high as 90% can be achieved. #OXB - George Pamenter, Maria Kapanidou, Ciaran Lamont, Lee Davies, Carol Knevelman & Kyriacos Mitrophanous.
