Neuroimmune biology of herpesvirus infections (NIMBUS) My long-term mission is to contribute to the prevention of herpesvirus-associated neurological and ocular disorders through research and education. Research activities will be focused on deciphering the interaction between pathogen and host factors in patients and healthy individuals that determine protection and pathogenesis of herpesvirus-associated neurological and indirectly ocular disorders in healthy and diseased human nervous tissues in-situ, ex-vivo and in-vitro. These studies are warranted to develop novel strategies leading to prediction (e.g. genotype virus and host) and early detection of disease (e.g. viral genome and biomarkers), avoiding worsening or recrudescent disease (e.g. antivirals and biological response modifiers) and ultimately prevention of herpesvirus-induced neurological and ocular disorders (e.g. vaccines). These issues will be addressed according to the NIMBUS concept using an interdisciplinary approach integrating knowledge in Virology, Immunology and Neuroscience at the dept. Viroscience and collaborating departments at the Erasmus MC. Additionally, educational activities are aimed to increase awareness of the aforementioned medical and scientific challenges by interaction and collaboration with stakeholders including patients, professionals and students.

Happy Monday! It's time for the "Invention of the Week"! Today we are going to be discussing the "TruHuX", developed by scientists at The University of Texas Health Science Center, San Antonio. 🐭 Because of their tiny size, ease of handling, similar immune systems and biological characteristics to humans, and sensitivity to genetic manipulation, mice are widely used in a variety of biological and biomedical research projects. TruHuX is a humanised mouse that has a complete and functional human immune system. According to the researchers, the model contains human T and B lymphocytes, memory B lymphocytes, germinal centers, thymus human epithelial cells, lymph nodes, and plasma cells that create highly specific autoantibodies and antibodies that are identical to those observed in humans. “By critically leveraging estrogen activity to support human stem cell and human immune cell differentiation and antibody responses, THX mice provide a platform for human immune system studies, development of human vaccines and testing of therapeutics,” Paolo Casali, MD, University of Texas Ashbel Smith Professor and Distinguished Research Professor. This novel finding offers up new avenues for human in vivo research, such as the creation of human bacterial and viral vaccinations, immunotherapeutics like cancer checkpoint inhibitors, and disease modeling. This innovative method has the potential to make non-human primates unnecessary for use in immunological and microbiological biomedical research. Dubbed TruHuX (THX), these humanized mice not only possess a human immune system but also harbor a human-like gut microbiome capable of generating specific antibody responses. This development marks a pioneering step towards advancing immunological and microbiological research, potentially offering a viable alternative to using non-human primates in these fields. The THX mice exhibit a range of human immune components, including lymph nodes, germinal centers, thymus human epithelial cells, and various types of human immune cells such as T and B lymphocytes, memory B lymphocytes, and plasma cells that produce specific antibodies and autoantibodies similar to those in humans. According to Paolo Casali, MD, a key figure in the research, these mice leverage estrogen activity to support the differentiation of human stem cells and immune cells, facilitating the development of human-like immune responses. This breakthrough opens up new avenues for human in vivo experimentation, including the study of human immune system functions, the development of human vaccines, and testing of therapeutics. 📣 #joboffer: Starting from 01.05.2024 for three years the BIH is looking for a Ph.D. student (f/m/d) - Translational Immunology.   The Sawitzki lab at the Berlin Institute of Health at Charité – Universitätsmedizin Berlin investigates the molecular mechanisms of immune cell activation and regulation, in order to better understand why components of the immune response themselves become a trigger of destructive inflammatory reactions, like e.g. in the context of autoimmune or COVID19 diseases. We are using especially multi-parametric technologies (cytometry, imaging, scRNAseq) to uncover pathological changes in the composition and function of immune cells in patient cohorts or clinical trials. The candidate will be involved in a project that aims to understand i) why certain patients develop severe immunopathologies in case of e.g. COVID-19 breakthrough infections and ii) whether or how the immune response differs from the immune response at initial infection.
