A paper has been published in which the rat BBB Kit is used! This study demonstrated that the poliovirus capsid protein VP3 can cross the vascular endothelial cells. two independent parts of VP3 contribute to this function and can enter human umbilical cord vascular endothelial cells and peripheral blood mononuclear cells. the VP3-derived peptide crossed the rat-derived blood brain barrier (BBB) model (BBB Kit™, PharmaCo-Cell, Japan) and migration from the blood side to the brain side was observed. These results provide new insights into the molecular mechanisms by which polioviruses invade the central nervous system (CNS). The BBB Kit is a breakthrough in vitro BBB model developed by PharmCo-Cell, which reconstitutes a BBB by co-culturing three types of BBB-related primary culture cells. Membranes rearranged, a fortress made, Protecting RNA, in their shade. In the ER and Golgi’s embrace, Virions assemble, with stealth and grace. Through the secretory pathway, they roam, Released into the bloodstream, to find a new home. Host factors intertwine, in a silent design, As HCV exploits, the dance of virus and host entwined. But when protease’s cleaver keen is blocked, The virus’s grip, by inhibition, is shocked. Without its slices, polyproteins remain whole, Viral replication halted, under control. Initiator Cell Death Induced by SARS-CoV-2 in Human Airway Epithelium SARS-CoV-2 triggers multiple cell death pathways, contributing to disease severity. Key findings from Liang et al. include: Primary Mechanism: Necroptosis was identified as the primary cell death mechanism, initiated by viral Z-RNA binding to Z-DNA binding protein-1 (ZBP-1) in ACE2-expressing cells and primary human airway epithelial (HAE) cultures. Apoptosis: Both infected and bystander cells underwent apoptosis, though it was primarily a bystander effect. Variant Comparison: The Delta variant induced significantly more Z-RNA/ZBP-1 interactions and necroptosis than Omicron, correlating with greater disease severity.
