A brilliant method for high-/-super resolution imaging of spheroids was just published in the Journal of Microscopy 📃🔬 This work is a collaboration between the groups of Dr. Susan Cox and Prof. Maddy Parsons, who is the director of the Nikon Imaging Center at King's College London! Very exciting to see the involvement of Rob Tetley from Nikon in this research! Harnessing the power of UV photolithography, the authors fabricate a polydimethylsiloxane stamp that is then used to create collagen well-like structures. These wells can nest spheroids with high spatial precision allowing for consistent high-resolution imaging. The authors validate the approach by visualizing a plethora of tumor spheroid behaviors. These include standard invasion assays imaged with the Nikon A1R Ti confocal as well as super-resolution filopodia activity at the spheroid periphery captured with the Nikon Ti2 Spinning Disc microscope. Accurate monitoring of orthotopic tumors in oncological research is critical. Magnetic resonance imaging (MRI) has been introduced as a modality to overcome limitations with common methods such as bioluminescence imaging (BLI).
