Sandra Czech, PhD student in our Department Mechanistic Toxicology, won a TEER measuring device, that was awarded by the PETA Sciene Consortium International. Congratulations 🤩🚀🥳! She was awarded for the project she is working on as part of her doctoral thesis. The project will focus on the development of new-approach methodologies (#NAM-based methods) for risk assessment of inhalation toxicants. Sandra explains 👩🔬💬: “To mimic the in vivo environment of the lung epithelium, we use primary normal human bronchial epithelial cells (#NHBE) cultured by using air-liquid interface (ALI) technology. Differentiated NHBE cultures will be exposed to typical lung toxicants and transcriptional responses will be analysed. For data analysis, we will move away from single gene comparisons and exploit co-expression and cross-process network analysis of toxicogenomic data. For selected chemicals, we will also measure the transepithelial uptake across NHBE barrier cultures and use these results as input parameter for our inhalation specific #PBPK model.” The new TEER equipment will be used to assess the epithelial barrier function and to implement data on the tightness of NHBE cultures as an indicator for barrier functionality and proper differentiation. This will be particularly important for the planned uptake and transport studies. In addition, TEER will be used as a readout for cell viability.
