I formed CNTRL Ventures to lead a round into Aanika… While pitching VCs on Aanika Biosciences, Inc. over this past fall, we started gaining incredible traction with customers who essentially control the global ag system. Just massive companies coming to us because of we have a proprietary tech that allows to write risk in new ways. So much so, that at one point when I was laying out our value proposition to a potential investor I thought: “Holy $#it, this is going to be massive…and it’s working”. I think pitch decks are total BS. I think companies need to CHEAPLY try a bunch of things to figure out what works and then go all in. When VCs debate market size it’s silly - everything is so non-linear you just can’t predict. You need to find crazy asymmetric opportunities. All this to say, I RE-sold myself hard on Aanika. We have guys on our cap table that have built multiple Billion dollar + startups in the ag space and exited, and very smart people who manage tens of billions in AUM across sectors. EVERY silicon valley VC we spoke to had nowhere near the domain expertise we have and therefore would just be a check and some good branding. Given that and the fact and that Aanika could be profitable next year, I had to change the plan. And thats why I formed Cntrl Ventures.. We are leading a round in Aanika becsuse I see massive upside Maybe at some point down (very very down) the road we’d invest in other companies and if we did there are a few ways I’d run it: 1. We’d be very controlling over the companies we invested in, like incredibly controlling and force commercializing their idea from day one.Id run it more like a prop trading firm where we allocate resources to highly talented people rather waste everyone’s time on pitch decks and “narratives”. 2. we would never ghost anyone. Just say you think the idea sucks but be direct about it. Hedge fund people much better in this respect because a one word email like “pass” says eveything. 3. Id never utter the words “be helpful”. It makes me want to 🤮. 4. No saving the world BS. You can’t make the world a better place if you never bring your thing to market. To do that is requires getting your hand dirty not networking. 5. Being seriously imbalanced would probably also be a requisite.

How do we minimize the number of protocol amendments and increase the operational feasibility of our clinical trials? This article below points out the benefits of using data, conversations and real world data to enhance the feasibility of the protocol design and inclusion/exclusion criteria: 1) Are we being too strict on our criteria in order to "control" for data variability? 2) Do are criteria allow for the intended patient population in the real world (if not, they need to) 3) How realistic is your design in how the study sites actually work? Since our last DataFAIR, there was been an overwhelming activities at the intersection of life sciences and GenAI. The Nobel Prize committee's decision aside, there has been ~1000s of papers published, ~100s of Foundation Models released, ~10s of companies funded all in the last 12 months. It is often quite hard to keep up and make sense of all of this. As we approach #datafair2024 on October 17 In Boston, we are taking our role as a credible AI-partner for our customers more seriously than ever. This year’s focus on #FoundationModels in biopharma presents significant opportunities for innovation which brings me to the key themes for this symposium: 1. I wonder if change in a mature industry is incremental or disruptive? I've been on the road last 3 weeks. One of my realizations is that the pharma/biotech industry is having a really hard time understanding the implications of AI in their field. Our conversations are centered on how AI might augment our old way of doing things. Not so much on will there be newer ways of doing things? Biological research is a complex balancing exercise between mechanistic understanding and pure empirical evidence. We are progressively better at understanding - or as I would say hypothesising - about how a disease works and how a drug might 'cure' it. A submission of mechanism of action is more common in IND applications but as far as I can tell it's not a pre-requisite. The greatest successes of drug research are solid empirical hit and trial. Very successful ones at that. The hay day of drug research in the 60s to 90s was because big pharma figured out how to do compound screening effectively at scale. Will AI give us new ways to do this in more effective ways? I would be extremely surprised if that were not to be the case. a. we have never had access to the kind of data that we do now b. we have never had a tool like modern AI to sift through data Those two combined will give us ways to do empirical research like we haven't done before. That much is clear. Will it change the relative dearth of blockbuster drugs? That is a whole another subject. What is clear to me is that the current focus on mechanistic understanding will have to evolve to incorporate AI. We will have newer ways to do things - not necessarily change or abandon old ways. “From Incubation to Acquisition: The growth Journey of an Early-Stage Company” - the story of Serotiny from inception through the turbulations of a biotech startup to an acquisition by Johnson & Johnson Innovative Medicine to bring their technology for oncology patients. Session led by Melinda Richter Global Head Johnson & Johnson innovation - JLABS, with Serotiny co-founder Colin F. (Also present is other co-founder Justin Farlow), Eric Moessinger Matthew Lorenzi and Leo MacDonald Serotiny started at JLABS section of the MBC incubator and through this connection got access to Johnson & Johnson experts who got excited despite the early stage and willingly joined the journey, shared insights about the killer experiments that needed to be done to get the data package for validation and helped in multiple other ways. Serotiny’s technology held the promise to allow Johnson & Johnson Innovative Medicine to tailor the design of cell therapy with diverse cell types. Multiple collaborations followed over several years to problem solve together, working closely as a team (“impossible to tell who is from Serotiny and who is from J&J), and an investment by J&J’d venture arm #JJDC. A strong and deep cultural fit became apparent in addition to the potential benefit of the technology, eventually leading to an acquisition just before new year’s 2023 aligned with the original vision of making the most impact through J&J’s big corporate engine and financial power. Post-acquisition and celebration, Serotiny remains in JLABS in the Bay Area. The team is key together, working on the same projects, keeping the same culture and focus on the science and plugged into the larger J&J organization. From an investor perspective (Eric Moessinger): excellent outcome. Colin’s learnings: talk to potential partners earlier and more. JLABS environment was a valuable resource - attended many programs in both Bay Area and DC JLABS locations to learn as much as possible about what’s awaiting downstream. Advice to startups: - See the many “no”s you’ll hear as opportunities to learn, polish your proposition, avoid mistakes. - be open about issues and problems. The right partner will see it as a sign of seriousness and humility - a recipe for success, rather than a liability, and roll up their sleeves to problem-solve. - articulate the differentiation of your company from all others. #mycompany is proud of its offering to innovators that attempts to respond to the needs of young startups: incubation and access to expertise, strategic venture funding at any stage that is aligned with desire to bring products to patients, ability to enter into early stage collaborations and licensing facilitated by a local team that is focused on building long term relationships and appreciates the challenges faced by our partners.

I'm delighted to share news of this new AI-focused drug discovery company, Xaira, built on foundational technology from David Baker and led by Marc Tessier-Lavigne. The company applies AI tools to unlock opportunities in drug discovery for aspects of biology that we believe are not readily obtainable with conventional methods. I co-led NEA's investment in Xaira, along with my NEA partner and the firm’s co-CEO Mohamad Makhzoumi, and I will be joining Xaira’s board. Our enthusiasm for Xaira based on our belief that applying AI and large language models to biology holds immense promise and is still in its early stages. Currently, the application of AI to biology relies on model architectures borrowed from the tech world. A lack of well-curated clinical training sets hinders building and training these models effectively. This limits the advancement of AI in solving biological problems compared to other fields. Xaira aims to bridge this gap. We now have a clear understanding of the data needed to answer fundamental biological questions. By iterating on leading life science models and using focused perturbation data generated through our large-scale experiments, Xaira will generate data specifically suited to building advanced model architectures that are optimized for biology -- and hopefully, unlock therapeutic opportunities for patients who are not being optimally served by existing therapeutics.
