So excited to share our recent publication on the groundbreaking advancements in human #mesenchymal stromal cells (hMSCs) research for manufacturing of #extracellular vesicles (EVs). Over the past 20 years, hMSCs have been pivotal in cell therapy, primarily due to their paracrine signalling mechanisms. Our study breaks new ground by demonstrating that human #telomerase reverse transcriptase (hTERT) immortalised #hMSCs can be adapted to suspension culture, eliminating the need for #microcarriers or other matrices to support cell growth. Key Highlights 🏄‍♂️ 🌷 Novel Cell Line: Introducing suspension hMSCs (S-hMSCs) with a doubling time of approximately 55 hours. 🌷Immunophenotype: Retains nearly 90% of CD73 and CD105 expression levels, with notable upregulation of CD44, CD46, and CD47. 🌷EV Production: Generates extracellular vesicles (EVs) in a chemically defined medium, averaging 150 nm in size, displaying key markers like CD63, CD81, and TSG101. 🌷Process Development: S-hMSCs offer immense potential for drug discovery and translational studies in the EV field. Last week, during a visit to our lab at Kura Biotech with some friends (Catalina del Río, Pamela Salazar Cifuentes, Raimundo Steidle Alcaíno, Angel Izurieta, Oliver Siegel & Matias Zegers), we discussed how similar protein expression was to software distribution, and how synthetic biology basically was about programming life instead of computers or robots. This article elaborates about some promising advances in the field of #DNA_Computers, how they work, and their impressive potential advantages, like parallel processing for game theory simulation and unparalleled storage capability (YouTube completely can be stored in 1 cubic centimeter of DNA). Check out our recent review article in #NatureReviewsNeuroscience A new era in CNS disorder treatment is dawning with gene therapy. Promising results in Spinal Muscular Atrophy (SMA), Amyotrophic Lateral Sclerosis (ALS), and Cerebral Adrenoleukodystrophy (CALD) offer the potential for improved quality of life for patients. Despite these successes, many gene therapy attempts haven't reached patients. Addressing delivery challenges and tailoring gene targets to specific pathologies is essential. Our latest review explores gene therapy modalities for CNS diseases, delivery strategies, and critical gaps in understanding. We also share lessons from failed trials to guide future developments.
