Last week, the URI Pharmaceutical Development Institute delivered real-world, hands-on industry training to Amgen employees in another customized training course entitled Cell Harvesting. Workshops included acid precipitation, centrifugation, and depth filtration exercises, where trainees aliquoted CHO cells into conical tubes, evaluated the impact of acid on the cells, used a bench-top centrifuge to remove cell debris and impurities from the cell culture, clarified a mixture of different sizes of beads to visualize how a depth filter separates larger components from smaller components, and evaluated cell changes during cell harvesting operations using light microscopy. It was another exciting week at the RI Biotech Boot Camp for the 20 trainees! Erin Bastien, Senior Analytical Scientist of the URI Pharmaceutical Development Institute, demonstrated how to isolate a valuable biomolecule from a complex biological system using the analogy of a strawberry extraction while Brian D. Plouffe, PhD, Assistant Professor in the Department of Cell and Molecular Biology (CMB) and academic advisor for the biotechnology program, described bioreactors and how filtration is key in biopharma to separate the biomolecule from other components of the cell. Taught by PDI and CMB's industry professionals, the DLT-funded boot camp covers real-world biotech industry topics including lab skills, current good manufacturing practices (cGMP), biopharma manufacturing concepts, aseptic operations, quality control systems, cleaning and environmental monitoring, and measurement and data collection. This week, the URI Pharmaceutical Development Institute kicked off the RI Biotech Boot Camp to provide hands-on, immersive workshop training to 20 trainees. To demonstrate the importance of aseptic handling in biopharma, Jennifer Lewis, Director of the URI Pharmaceutical Development Institute, led trainees in an activity to simulate a sterile fill-finish operation with green fluorescent protein (GFP). As Rhode Island Department of Labor & Training Director Matthew Weldon stated on day one of the 10-day boot camp, "this is a great opportunity. I'm challenging you: pay attention, show up, take advantage of everything they offer you." Today, the URI Pharmaceutical Development Institute welcomed back Amgen employees for another customized Advanced Cell Culture training workshop. Employees were trained in industry-focused, hands-on workshops, including cell line development, mammalian cell culture, analytical testing for critical control parameters, mAb production, and FDA/EMA/ICH regulatory compliance. The URI Pharmaceutical Development Institute deliver hands-on training to Amgen employees using their state-of-the-art facilities, equipment, and instrumentation, including a KAYE validator. Last week, trainees learned how perform a thermal mapping of an incubator -- from wiring thermocouples to calibration to PQ mapping to thermocouple verification. With active engagement in hands-on exercises, the participants gained a solid understanding of the importance of equipment qualifications including IQ, OQ, and PQ.
