🚨 New Research Spotlight 🚨 Another remarkable customer publication in Nature Cancer using 36-plex PhenoCycler (formerly CODEX) data from 401 patients with hepatocellular carcinoma (HCC) unveils a comprehensive spatial map of single-cell phenotypes and multicellular neighborhoods, shedding light on spatial heterogeneity in cancer progression. 🔬 Key Findings: 1. Spatial Niches: Tumor cells, CD4+ and CD8+ T cells, and macrophages cluster into distinct spatial niches, revealing how specific environments drive tumor behavior and progression. 2. Clinical Stratification: Researchers identified three distinct Spatial Phenotypes (SP-HI, SP-LI, and SP-PF), which could stratify clinical outcomes. SP-HI patients may benefit from immune checkpoint blockade (ICB), while SP-PF and SP-LI suggest alternative therapies such as tyrosine kinase inhibitors. 3. “The ICR is a leading institution for the discovery of new cancer drugs. Its researchers’ combination of expertise and knowledge will certainly lead to new and better treatments for children with cancer.” Andrea Lampis is a Higher Scientific Officer in the Paediatric Tumour Biology group, led by Professor Louis Chesler, at the ICR. He coordinates spatial multiplexed imaging for the team, capturing detailed images of biological samples. He says that a typical day involves working on experiments with his team and analysing and reviewing data. He is currently involved with research projects into childhood cancers including neuroblastoma and medulloblastoma. “The ICR is an internationally recognised institution in the study of childhood cancers. It has many teams passionately dedicated to understanding the biology of these complex and devastating diseases, in order to better treat them. “We are fortunate that we have financial support in the form of funding from organisations such as the TeamLuke Foundation set up in memory of Luke Bell who sadly passed away aged eight from neuroblastoma in 2018. 🚨 New Research Spotlight 🚨 Featured on the cover of Nature Cancer, this groundbreaking study unravels the immune biology of immune-related adverse events (irAEs) caused by immune checkpoint inhibitors (ICIs) and proposes potential treatment strategies beyond corticosteroids. 🔬 Key Findings: 1. Multiplex immunohistochemistry (mIHC) was crucial in identifying increased IL-17A expressing CD4+ T cells at the site of irAE inflammation in conditions like skin rash and colitis, providing a deep, spatial understanding of immune responses. 2. This aligns with serum proteomic analyses that revealed aberrant T cell activity and type I & III immune signatures, particularly in severe irAEs. Blocking IL-17A in patients with severe, corticosteroid-refractory irAEs led to a successful resolution of symptoms. 3. I #TRONpublications I We are proud to share our recent publication entitled “Temporal evolution and inter-patient heterogeneity in primary and recurrent head and neck squamous cell carcinoma” - the result of a longstanding collaboration with the Institute of Head and Neck Studies (InHANSE), Birmingham, UK. Head and neck squamous cell carcinomas (#HNSCCs) are heterogeneous tumors with a high certainty of genetic modifications from initial diagnosis to recurrence. This can have dramatic implications on the selection of appropriate therapies, especially targeted #immunotherpapies. Here, the team systematically analyzed whole exome and transciptome sequencing data of paired primary and recurrent samples from a cohorts of nine #HNSCC patients 🧬👩💻. The results strengthen the importance of close monitoring of genetic changes during treatment and disease progression and advocate a re-biopsy in the event of recurrence and treatment failure, in order to select the most appropriate therapeutic strategy.
