    💡 #DPinitol promotes tau dephosphorylation through a cyclin-dependent kinase 5 regulation mechanism: A new potential approach for #tauopathies? Recent evidence links brain insulin resistance with neurodegenerative diseases, where hyperphosphorylated tau protein contributes to neuronal cell death. In our recent study, we aimed to evaluate if d-pinitol inositol, which acts as an insulin sensitizer, affects the phosphorylation status of tau protein. 🔬 We studied the pharmacological effect of d-pinitol on insulin signaling and tau phosphorylation in the hippocampus of Wistar and Zucker rats.  Surprisingly, we discovered that oral d-pinitol treatment lowered tau phosphorylation significantly, but not through the expected kinase GSK-3 regulation. An extensive search for additional kinases phosphorylating tau revealed that this effect was mediated through a mechanism dependent on the reduction of the activity of the CDK5, affecting both its p35 and p25 subunits.  The present findings suggest that d-pinitol, by regulating CDK5 activity through a decrease of CDK5R1, is a potential drug for developing treatments for neurological disorders such as tauopathies. 🔈 The British Journal of Pharmacology, one of the most prestigious European scientific journals covered our key findings: 1️⃣ D-pinitol (prresent in our marketed brand Agelity) reduces tau protein phosphorylation in the hippocampus, a brain area controlling memory management. It does so through a mechanism involving the CDK5 kinase and its receptor, the protein p35/p25. 2️⃣ This is a unique mechanism for controlling the deposit in the brain of tau protein, a crucial event in the pathogenesis of neurodegenerative disorders such as tauopathies, which include Alzheimer’s Disease and other dementias. 3️⃣ D-pinitol effects on tau phosphorylation were also observed in humanized mouse models of Alzheimer’s disease, the so-called 3-TG mouse. 4️⃣ The effects were observed by administering Agelity through the oral route (in the drinking water). 5️⃣ Advenion has patented Ageliyt as a potential treatment for tauopathies, such as frontotemporal dementia, Pick Disease, Progressive supranuclear palsy, or chronic traumatic brain injury.

    Dietary administration of D-chiro-inositol attenuates sex-specific metabolic imbalances in the 5xFAD mouse model of #Alzheimer's disease Increasing evidence shows that hypothalamic dysfunction, insulin resistance, and weight loss precede and progress along with the cognitive decline in sporadic Alzheimer's Disease (AD) with sex differences. One of our studies aimed to determine the effect of oral dietary administration of D-Chiro-inositol (DCI), an inositol used against insulin resistance associated with polycystic ovary, on the occurrence of metabolic disorders in the transgenic 5xFAD mouse model of AD (FAD: Family Alzheimer's Disease).  Energy balance and multiple metabolic and inflammatory parameters in the hypothalamus, liver, and plasma were evaluated to assess the central and peripheral effects of DCI. Results indicated that weight loss and reduced food intake in 5xFAD mice were associated with decreased neuropeptides controlling food intake and the appearance of a pro-inflammatory state in the hypothalamus. Our study depicts that metabolic impairment along with the cognitive decline in a mouse model of AD, which is exacerbated in females, can be ameliorated by oral supplementation with insulin-sensitizing DCI. This study, published in Biomedicine & Pharmacotherapy, is the result of collaboration with FIMABIS (Andalusian Public Foundation for Health and Biomedicine Research in Malaga), describes the pharmacological efficacy of D-chiro-inositol to restore the metabolic balance lost as results of Alzheimer’s Disease (AD). This article demonstrates the following: 1️⃣ A humanized 5XFAD mouse model of Alzheimer’s disease exhibits nutritional and metabolic alterations often found in human patients. Administration of D-chiro-inositol counteracts these metabolic alterations, without the need for additional myoinositol supplementation. 2️⃣  D-chiro-inositol counteracts the alterations in weight loss, energy expenditure, hypothalamic inflammation, and circulation insulin/GIP/GLP-1 found in the mouse model of Alzheimer’s disease. 3️⃣ D-chiro-inositol is more efficient in female 5XFAD animals, counteracting the development of fatty liver and favoring fatty liver oxidation. 4️⃣ The metabolic improvement resulting from D-chiro-inositol administration is achieved through the oral route (administration in the drinking water), and it is accompanied by cognitive enhancement, supporting its use as a medical food for healthy aging, especially in women. 5️⃣ Euronutra has patented Gyneos as a potential treatment for tauopathies-associated damage, opening a new era of nutraceutical support for aging and neurodegeneration.
