Sucrose

Leptin receptors are present in taste buds and previous research indicates that leptin administration modified electrophysiological and behavioral responses to sweet taste. It is now known that sweet taste is temperature dependent. We examined the influence of (1) stimulus temperature on chorda tympani (CT) nerve responses to sucrose, saccharin and NH(4)Cl; and (2) leptin administration on CT nerve responses to sucrose, saccharin and other basic taste stimuli at 35°C that maximized sweet-taste sensitivity in C57BL/6 mice. We found that the CT nerve responded with greater magnitude to sucrose and saccharin as stimulus temperature increased from 23 to 35°C and then declined at higher temperatures. In contrast, the CT nerve responses to NH(4)Cl increased in magnitude as temperature increased from 23 to 44°C. We also showed that leptin selectively increased the CT nerve responses to sucrose at 35°C in both fasted and free-fed mice. The responses of mice treated with the saline vehicle did not change. Our findings are consistent with the notion that leptin binds with its receptors in fungiform taste buds and alters the message conveyed by sugar-responsive neurons to the brain.

Methods for accurate and efficient quantification of metabolic fluxes are desirable in plant metabolic engineering and systems biology. Toward this objective, we introduce the application of "bondomers", a computationally efficient and intuitively appealing alternative to the commonly used isotopomer concept, to flux evaluation in plants, by using Catharanthus roseus hairy roots as a model system. We cultured the hairy roots on (5% w/w U-(13)C, 95% w/w naturally abundant) sucrose, and acquired two-dimensional [(13)C, (1)H] and [(1)H, (1)H] NMR spectra of hydrolyzed aqueous extract from the hairy roots. Analysis of these spectra yielded a data set of 116 bondomers of beta-glucans and proteinogenic amino acids from the hairy roots. Fluxes were evaluated from the bondomer data by using comprehensive bondomer balancing. We identified most fluxes in a three-compartmental model of central carbon metabolism with good precision. We observed parallel pentose phosphate pathways in the cytosol and the plastid with significantly different fluxes. The anaplerotic fluxes between phosphoenolpyruvate and oxaloacetate in the cytosol and between malate and pyruvate in the mitochondrion were relatively high (60.1+/-2.5 mol per 100 mol sucrose uptake, or 22.5+/-0.5 mol per 100 mol mitochondrial pyruvate dehydrogenase flux). The development of a comprehensive flux analysis tool for this plant hairy root system is expected to be valuable in assessing the metabolic impact of genetic or environmental changes, and this methodology can be extended to other plant systems.

To determine the pH and sucrose concentrations (SC) of pediatric liquid drugs of long-term use by children in order to evaluate the potential risk for dental caries and dental erosion. After assessing the pH, we analyzed 71 aqueous medicine samples for sucrose by the Lane-Eynon general volumetric method. The pH and SC values (mean +/- standard deviation (SD) were calculated according to therapeutic action. The highest and the lowest SC values (mean +/- SD) were found in respiratory (37.75% +/- 17.23%) and endocrine drugs (11.97% +/- 15.16%) (p < 0.01). The values for medicines prescribed for daily ingestion were 47.15% +/- 9.57%, whereas for twice daily and three or four times a day, these numbers were 24.42% +/- 18.03% and 34.43% +/- 14.83%, respectively (p < 0.01). The SC (mean +/- SD)) values were higher in syrups (36.32% +/- 17.62%) than in other formulations (p > 0.05). The overall pH (mean +/- SD) was 5.89 +/- 2.02 (range 2.3 +/- 0.01 to 10.6 +/- 0.02). In products with acidic pH, the SC (mean +/- SD) was significantly lower (22.14% +/- 15.72%) than in nonacidic medicines (39.22% +/- 15.82%) (p < 0.001). It can be concluded that the pediatric medicines studied have a high SC and low pH, which vary according to therapeutic class, daily dose, and brand. Caution about dental caries, dental erosion, and systemic diseases such as diabetes mellitus is warranted when these medicines are ingested frequently.

An experiment was conducted to find out the suitable preservative solution which provides the longest vase life for yellow gladiolus against eight preservative solutions coded from T 0 to T 7 and these were T 1 : 100-ppm silvar thiosulphate (STS); T 2 : 3% sucrose + 100-ppm citric acid (CA); T 3 : 3% sucrose + 100-ppm salicylic acid (SA); T 4 : 3% sucrose + 100-ppm STS; T 5 : 5% sucrose + 100-ppm CA; T 6 : 5% sucrose + 100-ppm SA; T 7 : 5% sucrose + 100-ppm STS with T 0 : Control (Tap water). T 4 treatment provided maximum spike diameter (13.9 mm) and floret opening (96.8%) at 12 th days after treating, maximum days to basal floret senescence (12.4), maximum no. of floret open during basal floret senescence (10.5), maximum solution absorption (47.8 ml/spike), maximum vase life (21.5 days) with minimum fresh weight loss (25.1%) and floret wilting (29.3%) at 12 th days after treating which was closely followed by T 3. However, incidence of stem rotting was not in T 2 , T 3 and T 4. Blend solutions of STS/SA (100-ppm) and 3% sucrose is recommended to use for the maximization of the postharvest life of yellow gladiolus.
