This article presents the results of the application of molecular genetic research methods in the analysis of the genus Hedysarum L. in the flora of the North Caucasus.Materials and methods. The function of the phosphorylation of histone H3 at Ser 10 in plant cell division is uncertain. ... more The function of the phosphorylation of histone H3 at Ser 10 in plant cell division is uncertain. The timing correlates with chromosome condensation, and studies in plant meiosis suggest that it is involved in sister chromatid cohesion. In mitosis, plant chromosomes are highly phosphorylated in the pericentromeric region only. In order to modulate H3 phosphorylation, root meristems of different plant species were treated with the protein phosphatase inhibitor cantharidin or with ice-water. Immunostaining using an antibody specific to phosphorylated H3 at Ser 10 revealed a high level of H3 phosphorylation along the whole mitotic chromosome after cantharidin treatment, which resembles the distribution seen exclusively in first meiotic division. In chromosomes that were isolated from meristems treated with ice-water, the heterochromatic regions and nucleolar organizer regions, in addition to the pericentromeric region, were highly phosphorylated at H3. Two types of soils were studied: contaminated one collected near of copper-smelters (the Middle Urals) and Ni-enriched soil from an area of a natural geochemical anomaly (the Polar Urals). We measured potentially mobile forms of metals in soils and water-soluble concentrations of metals in soils with standard methods. Allium roots were exposed in aqueous soil extracts for 30 hours with a recovery duration for 20 and 44 hours. We found an increase of frequencies of total nuclear anomalies in meristematic cells as compared to control samples. The most frequent types of anomalies in all studied samples were extrusions. Influence of high Ni concentrations leads to decrease of roots growth that was observed after recovery time also and increasing of number of micronucleus. Rosa × archipelagica is a spontaneous hybrid between Rosa rugosa and R. maximowicziana distribute... more Rosa × archipelagica is a spontaneous hybrid between Rosa rugosa and R. maximowicziana distributed, together with its parental taxa, on the coast of a small Stenina Island in the Peter the Great Gulf, the Sea of Japan (Russian Far East). Its hybrid origin is confirmed by direct electropherogram comparisons of PCR products and maximum likelihood analyses of nuclear ITS 1–2 and plastid intergenic spacers trnH–psbA and ndhC–trnV. The plastome markers indicate R. maximowicziana as the maternal genome donor of Rosa × archipelagica, however, one specimen shows also signs of heteroplasmy. This is the first evidence of possible biparental inheritance of plastids in rose hybrids. The results do not show whether the hybrid plants all belong to F1 or further generations of hybrids.
