Viral infection in fruit flies can alter intestinal homeostasis and lead to physiological changes. Researchers from Institut Pasteur and Institut Curie in Paris employed fluorescently labeled #DNA #oligonucleotides printed by enzymatic DNA synthesis (#EDS) to track viral infection of the fruit fly gut using a technique called RNA FISH. The group found that infection of flies with the Drosophila A virus (DAV) led to changes in intestinal homeostasis, including an over-proliferation of intestinal stem cells (ISCs). The group found that DAV infection caused cytokine release through a JNK signaling pathway, leading to ISC proliferation through growth factor signaling pathways. Sting-Relish NF-κB signaling was found to be required for the ISC proliferation, and the over-proliferation of ISC cells within the fruit fly #gut led to a shortened #lifespan. A marker for age-associated intestinal dysplasia was upregulated in infected flies.
