Certain species of Mucorales have been identified as causative agents of mucormycosis, a rare yet often lethal fungal infection. Notably, these fungi exhibit intrinsic resistance to common azole drugs, which target lipids. Given the pivotal role of lipids in drug resistance and their contribution to innate resistance to azoles, this study provides a comprehensive overview of key lipid classes, including sphingolipids (SLs), glycerophospholipids (GPLs), and sterols, in Rhizopus delemar 99-880, a well-characterized reference strain among Mucorales. Using shotgun lipidomics as well as liquid- and gas-chromatography-based mass spectrometric analyses, we identified the lipid intermediates and elucidated the biosynthetic pathways of SLs, PGLs, and sterols. The acidic SLs were not found, probably because the acidic branch of the SL biosynthesis pathway terminates at α-hydroxy phytoceramides, as evident by their high abundance. Intermediates in the neutral SL pathway incorporated higher levels of 16:0 fatty acid compared to other pathogenic fungi. A strikingly high phosphatidylethanolamine (PE)/phosphatdylcholine (PC) ratio was observed among GPLs. Ergosterol remains the major sterol, similar to other fungi, and our analysis confirms the existence of alternate ergosterol biosynthesis pathways. The total lipidomic profile of R. delemar 99-880 offers insights into its lipid metabolism and potential implications for studying pathogenesis and drug resistance mechanisms. Full article

Nowadays, cotton boll detection techniques are becoming essential for weaving and textile industries based on the production of cotton. There are limited techniques developed to segment, detect, and count cotton bolls precisely. This analysis identified several limitations and issues with these techniques, including their complex structure, low performance, time complexity, poor quality data, and so on. A proposed technique was developed to overcome these issues and enhance the performance of the detection and counting of cotton bolls. Initially, data were gathered from the dataset, and a pre-processing stage was performed to enhance image quality. An adaptive Gaussian–Wiener filter (AGWF) was utilized to remove noise from the acquired images. Then, an improved Harris Hawks arithmetic optimization algorithm (IH2AOA) was used for segmentation. Finally, an anchor-free compact central attention cotton boll detection network (A-frC2AcbdN) was utilized for cotton boll detection and counting. The proposed technique utilized an annotated dataset extracted from weakly supervised cotton boll detection and counting, aiming to enhance the accuracy and efficiency in identifying and quantifying cotton bolls in the agricultural domain. The accuracy of the proposed technique was 94%, which is higher than that of other related techniques. Similarly, the precision, recall, F1-score, and specificity of the proposed technique were 93.8%, 92.99%, 93.48%, and 92.99%, respectively. Full article

Background/Objectives: The aim of this study was to evaluate the choroidal vascular response using optical coherence tomography (OCT) in patients with chronic central serous chorioretinopathy (CSCR) during transient increases in blood pressure. Methods: This observational, case–control study enrolled chronic CSCR patients and age-matched healthy controls. OCT scans of the macula were performed at rest and during hand-grip (HG) isometric exercise. Mean ocular perfusion pressure (MOPP) and subfoveal choroidal thickness (SCT) were measured at baseline and during stress. Quantitative OCT assessment included the bright area (BA, stroma), dark area (DA, vascular lumen), and total choroidal area (CA). The choroidal vascularity index (CVI) was calculated as DA/CA to assess vascular response to stress. A comparative analysis between CSCR patients and controls was conducted. Conclusions: MOPP was significantly higher (p = 0.008) at baseline in CSCR patients and further increased under stress compared to controls. SCT and CA were both significantly higher in CSCR patients than in healthy subjects at rest and under stress (p < 0.001), but no change occurred after HG. A significant decrease in CVI (p = 0.005) was noted in controls under stress, but not in CSCR patients. Additionally, a negative correlation between CVI and MOPP was found in healthy subjects (−0.648; p = 0.043). The study demonstrated a choroidal vasoconstrictive response to stress in healthy subjects, as evidenced by a decrease in CVI, but not in CSCR patients. This suggests that CSCR patients may experience impaired choroidal blood flow regulation, resulting in potentially higher perfusion pressures during stress without compensatory vasoconstriction, potentially affecting the choriocapillaris. Background/Objectives: The receptor binding motif (RBM) of the SARS-CoV-2 spike protein is critical for viral entry into host cells. Development of a vaccine targeting this region is a promising strategy for COVID-19 prevention. To enhance the immunogenicity of SARS-CoV-2 vaccines, we developed an adenoviral vector expressing the RBM from the SARS-CoV-2 spike protein that fused to the human Fc (hFc) domain. Methods: The recombinant RBM_hFc fusion protein was successfully cloned into the pacAd5CMV-N-pA (pAd5) vector and expressed in HEK293 cells as a ~40 kDa protein. A recombinant adenovirus encoding RBM_hFc was subsequently generated and confirmed by cytopathic effect assay. Results: Western blot analysis verified the expression of RBM_hFc in the adenovirus (AdV). ELISA assays, validated for IgG detection, demonstrated a twofold increase in IgG antibody levels (M–1.090 at 450 nm; SD—±0.326; and 95% CI—0.250 [0.839 to 1.340]) in sera from BALB/c mice immunized with Ad/RBM_hFc, compared to the negative control group. Result suggests a robust humoral immune response induced by the Ad/RBM_hFc vaccine. Moreover, ELISpot assays demonstrated a tenfold increase in IFN-γ -producing cells (M—440 spot-forming cells; SD—±124.976; and 95% CI—75.522 [364.478 to 515.522]) in mice immunized with AdV/RBM_hFc compared to the negative control group. Result proved that AdV/RBM_hFc-stimulated a robust cellular immune response in animal model. Conclusions: Our findings indicate that the RBM_hFc fusion protein enhances both humoral and cellular immune responses. These results suggest the potential of adenoviral vectors carrying RBM_hFc as vaccine candidates. However, comprehensive evaluation of the protective efficacy of these adenoviral vectors will necessitate rigorous experimental studies. Full article

Background: Controlled non-enzymatic glycation reactions are common under normal physiological conditions. However, during elevated blood glucose conditions, the glycation reactions are accelerated, leading to the formation of toxic compounds such as advanced glycation end products (AGEs). Several natural products are now being investigated as protective agents against glycation to preserve blood protein structure and functions. Methods: Human serum albumin (HSA) was glycated with 0.05 M α-D-glucose alone or in the presence of Zingiber officinale Roscoe (ginger) extract (0.781–100 μg/mL) for 10 weeks, and biochemical, biophysical, and computational analyses were carried out. Results: HSA glycated for 10 weeks (G-HSA-10W) resulted in significant production of ketoamines, carbonyl compounds, and AGE pentosidine. Notable structural alterations were observed in G-HSA-10W, ascertained by ultraviolet (UV), fluorescence, and circular dichroism (CD) studies. Antioxidant, anti-glycating, AGEs inhibitory, and antibacterial effects of ginger extracts were observed and attributed to the presence of various phytochemicals. Molecular docking studies suggested that the compounds 8-shagaol and gingerol exhibited strong and multiple interactions with HSA. Molecular simulation analysis suggests HSA attains a high degree of conformational stability with the compounds gingerol and 8-shogaol. Conclusions: These findings showed that ginger extract has an antioxidant function and can prevent glycation-induced biochemical and biophysical alterations in HSA. Thus, aqueous ginger extract can be utilized to combat glycation and AGE-related health issues, especially diabetes, neurological disorders, inflammatory diseases, etc. Full article

This study provides a detailed bibliometric analysis of research on medicinal plant extracts conducted over the past decade, with a focus on their therapeutic applications. Using the Scopus database, 1105 publications from 2014 to 2024 were analyzed to uncover trends in publication volume, collaboration networks, and research clusters. The analysis revealed a steady annual growth rate of 1.98%, with most studies concentrating on the anti-inflammatory, antimicrobial, and antioxidant properties of plant extracts, particularly those containing bioactive compounds such as flavonoids, alkaloids, and terpenoids. The study also highlighted the increasing international collaborations in the field, with 30.84% of the documents featuring cross-border co-authorship. Leading contributors to this research include India, China, and Italy, whose researchers and institutions have played a significant role in advancing the field. The widespread use of advanced analytical techniques, including High-Performance Liquid Chromatography (HPLC) and Nuclear Magnetic Resonance (NMR) spectroscopy, has enhanced the identification and characterization of bioactive molecules. Keywords such as “plant extract”, “antioxidant activity”, and “phytochemistry” were the most frequently occurring, reflecting dominant research themes. Additionally, the h-index analysis showed that 59 of the documents had received at least 59 citations, highlighting the significant impact of this body of research. The study underscores the growing role of phytochemicals in the development of alternative therapies for chronic diseases such as cancer, diabetes, and neurodegenerative disorders. These findings provide a roadmap for future research, encouraging further exploration of plant-based therapies with an emphasis on sustainability, bioactive compound efficacy, and global collaboration. Background: We report a case of brainstem anesthesia (BSA) and subsequent cardiac arrest following a routinary peribulbar block (PB) in a patient scheduled for cataract extraction and intraocular lens (IOL) implantation, thus providing a reference for further analysis of this potentially catastrophic life-threatening complication and to evaluate the current knowledge in terms of incidence, physiopathology management, and treatment of the BSA following PB. Methods: Three databases (PubMed, Embase, and Scopus) were checked to perform a systematic review of all available studies in the English Language following the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines to evaluate relevant studies that clearly described BSA following BSA. Results: Our literature search identified 15 cases. All the patients experienced BSA-related symptoms, including respiratory arrest, seizure, heart rate, and blood pressure abnormalities. All the patients with respiratory arrest required assisted ventilation with intubation, whereas patients with seizures were administered intravenous midazolam. Only one patient experienced cardiac arrest and underwent cardiac resuscitation. Surgery was aborted and deferred in 5 out of 15 patients, whereas 4 out of 15 underwent surgery after extubation. Conclusions: Despite the BSA incidence after the PB being very low, this possible life-threatening rare event should be considered in patients losing their consciousness and becoming apneic after the block. Therefore, prompt recognition and immediate treatment are paramount to cope with this potentially catastrophic scenario and save the patient’s life. Full article

The once-dominant view that architectural developments in mediaeval Southeast Asia closely followed Indian ‘influence’ is now largely rejected. Recent scholarship has shifted its focus onto the agency of local artists and architects in driving architectural innovations across the region. However, specific cases of transregional exchanges in architectural ideas and practices remain underexplored. This study examines three geographically distant Buddhist sites—Paharpur in northern Bangladesh, Candi Sewu in Central Java, Indonesia, and Samye Monastery in central Tibet—active in the late 8th century. I consider the significance of specific similarities and their temporal correlations within a broader range of styles, materials, and technologies. I argue that the activity at these sites reveals a shared architectural agenda transmitted over vast distances by religious experts, including Buddhist monks, in the last decades of the 8th century. Central to the network of three temple-monasteries proposed is the role that a specific architectural type was understood to play in protecting the kingdom and extending a king’s sovereignty while manifesting his spiritual aspirations. By distinguishing between architectural forms, architectural agendas, and modes of production, this study clarifies the complex nature of transregional architectural exchange in the premodern world. The present study investigates the synthesis and characterization of a zirconium oxide (ZrO2)-based metal composite doped with cerium (Ce) and yttrium (Y), using chromium (Cr) and nickel (Ni) as base metals. These constituents were selected for their superior mechanical properties and compatibility with the ceramic phase. High-purity powders were homogenized via high-energy ball milling, followed by cold pressing and sintering in a controlled atmosphere of hydrogen. The sintering process was conducted at temperatures ranging from 850 °C to 1350 °C to examine the evolution of microstructure, grain growth, and densification. Scanning electron microscopy (SEM) revealed a homogeneous distribution of phases, with distinct microstructural features attributed to each element at different sintering temperatures. The experimental results revealed that the composite’s density was increased by 30% and porosity was reduced by 61% at a sintering temperature of 1350 °C. The hardness and flexural strength of composite were found to be 23% and 60% higher at 1350 °C, respectively, compared to that at 850 °C, suggesting enhanced mechanical properties due to cerium and yttrium reinforcement within matrix and efficient doping and phase transformation. Overall, incorporation of cerium and yttrium significantly improved mechanical behavior and phase stability of ZrO2–Cr–Ni composite, highlighting its potential for advanced engineering applications. This paper systematically explores the applications of image processing techniques in machined surface analysis, a critical area in industries like manufacturing, aerospace, automotive, and healthcare. It examines the integration of image processing in traditional Computer Numerical Control (CNC) machining and micromachining, focusing on its role in tool wear analysis, workpiece detection, automatic CNC programming, and defect inspection. With AI and machine learning advancements, these technologies enhance defect detection, surface texture analysis, predictive maintenance, and quality optimization. The paper also discusses future advancements in high resolutions, 3D imaging, augmented reality, and Industry 4.0, highlighting their impact on productivity, precision, and challenges such as data privacy. In conclusion, image processing remains vital to improving manufacturing efficiency and quality control. Full article

Abstract: Aluminium and its composites are widely used in production to enhance the strength of lightweight objects. In this study, an AA7075/SiC composite was fabricated using a stir casting route. Multi-objective optimization and finite element analysis were performed with various process parameters on a manufactured aluminium composite (AA7075 + SiC) undergoing a laser beam welding process. Four welding parameters, i.e., pulse frequency, power, welding speed (transverse), and wire size were taken for laser welding as per the L-9 orthogonal array for experimental study. Tensile strength, deflection, temperature distribution, Rockwell hardness (fusion zone), and Rockwell hardness (heat affected zone) were taken as output parameters after welding. The standard deviation objective weighting–grey relational optimization method optimized the process parameter. ANSYS APDL 23 software was utilized to simulate the entire laser welding method with a cylindrical heat source to predict the temperature distribution in the butt-welded plates. This software uses finite element analysis and gives a deviation of only 5.85% for temperature distribution with experimental results. This study helps to understand the effect of various parameters on the welding strength of the aluminium composite. Full article
