The Fluid Management business unit of Asahi Kasei Bioprocess has launched DAC Ergo, a liquid chromatography column design that, according to the company, allows large-scale stainless steel dynamic axial compression LC columns to be safely unpacked, disassembled and reassembled by a single operator in minutes using a hydraulic system combined with a bottom plate fastening approach.
Constructed out of stainless steel to withstand the operating backpressures, the weight of these large-scale columns has historically resulted in a cumbersome experience for end users during column unpacking, disassembly and re-assembly. The DAC Ergo supports the growing demand for DAC columns that are required for large-scale cGMP purifications of microbial-derived proteins, oligonucleotides and peptides.
"DAC columns are used regularly to pack 10 to 40-micron particle size chromatography media for large-scale purifications of microbial-derived proteins and oligonucleotides," said Michael Brandt, Lead Innovation Engineer for Asahi Kasei Bioprocess America. "Our observation has shown that many pharmaceutical manufacturers schedule 3-4 operators for a full day shift to successfully complete the unpacking, disassembly and reassembly of legacy large stainless-steel chromatography columns. We decided not to accept that and set out to improve the safety profile and the user experience."
DAC Ergo is currently available in column diameters between 60 cm and 120 cm i.d. at pressure ratings up to 35 bar. Additional diameters and pressure ratings are in development.
