Its southern terminus at U.S. Highway 71 approximately {{convert|1|mi|km|adj=on}} south of [[Falls Chapel, Arkansas|Falls Chapel]]. It heads north through Nashville where soon after it merges with highway 26 which carries it into Murfreesboro, picking up highway 19 in Murfreesboro. Continuing north soon after it merges with U.S. 70 and Arkansas 84 which spilts off to the west in Salem, Arkansas. Highway 27 continues northeast following 70 into Glenwood where it jonis up with Highway 8 carrying it to Norman where 27 continues north once more to Mt. Ida. A short concurrency with U.S. 270 here before it heads north towards [[Dardanelle, Arkansas|Dardanelle]]. After picking up Highway 28 at Rover and Highway 10 at Danville. it continues north and ends at Highway 7 in Dardenelle. It begins again along Highway 7 at Dover and heads northeast to Hector where it turns north to Tilly where it merges with Highway 16. Soon after Tilly it turns southeast and enters Van Buren County before turning north to Marshall. From here it continues north to end in Harriet at Highway 14.
